PhD position (f/m/x) – Molecular Characterisation of a Pathogenic KCNJ5 Mutation and its Effect on the Adrenal Gland

for 36 months, and a salary in accordance with the TV-L pay scale.

Within the DFG-TRR205 collaborative research center, adrenal disorders are investigated through a highly interdisciplinary framework. Mutations in the KCNJ5 gene are among the most common causes of primary hyperaldosteronism in humans. In Project B19, we study a KCNJ5-mutant pig model that serves as a translational large-animal model for familial hyperaldosteronism type III (https://adrenal- research.de/project-b19/). This model provides a unique opportunity to bridge the gap between basic research and clinical application in this important human disease.

Your Role

In this project, you will investigate the impact of a pathogenic KCNJ5 mutation on adrenal gland function using a recently developed large-animal model. You will apply state-of-the-art methodologies, including omics approaches, with a focus on single-cell transcriptomics, in situ functional analyses of tissue slices, histology and advanced imaging, cell culture techniques and molecular biology methods. Your research will aim to define the molecular signatures and regulatory mechanisms that drive aldosterone-producing adrenal cells harbouring mutant KCNJ5 channels. In particular, you will explore how loss of ion selectivity in this transporter triggers the transition from a physiologically quiescent state to a pathogenic, autonomously aldosterone-producing phenotype.

  • An outstanding degree in veterinary medicine, biomedicine, molecular medicine, biochemistry, biology, or a related field
  • A strong passion for research, with high motivation and enthusiasm
  • Excellent communication skills and the ability to work both independently and collaboratively in an interdisciplinary team
  • A strong interest in data processing, analysis, and biostatistics
  • A keen interest in, and ideally experience with, hands-on laboratory work

Our laboratories are located at the Gene Center, the ICON, and the Center for Innovative Medical Models (CiMM) of LMU Munich, all of which provide state-of-the-art research infrastructure. Downtown Munich and the Bavarian lakes can be reached in 20min, the beautiful German Alps in under 1h. The position is funded for three years, with a salary according to the German public-sector pay scale (TV-L), depending on experience.
