Johannes Stigler's team reveals how the chromatin remodeler INO80 uses DNA shape cues and chromatin-bound factors to precisely position nucleosomes at promoters and replication origins, providing new insight into the mechanisms that shape genome organization.
Using single-molecule DNA curtain assays, the researchers discovered that INO80 actively scans DNA and is guided by both DNA shape features and chromatin-bound proteins. Together, these mechanisms help establish precise nucleosome patterns at key regulatory regions, ensuring proper control of gene expression and DNA replication
